In this study the involvement of the dorsolateral periaqueductal gray matter (dlPAG) and the caudate-putamen in the performance of conditioned taste aversion (CTA) and passive avoidance (PA) in the rat has been investigated. The ability of rats to perform both tasks under anaesthesia, or under ketamine and xylazine (KX) anaesthesia was assessed. Furthermore, the involvement of the dlPAG in the performance of the tasks was assessed by microinjection of muscimol and ibotenic acid into the dlPAG. In addition, for the latter experiment electrolytic lesions were applied bilaterally to the frontal pole of the caudate-putamen. In all experimental groups avoidance responses were observed in both tasks. Ketamine anaesthesia did not interfere with the performance of the tasks at doses up to 200 mg/kg. However, higher doses, although still allowing for the performance of the tasks, resulted in incoordination. The tolerance to the effects of KX was more pronounced in the CTA than in the PA task. Both muscimol and ibotenic acid interfered with the performance of the tasks in a dose-dependent manner. Electrolytic lesions of the dlPAG interfered with the performance of the CTA more than the PA task. The results show that anaesthesia does not interfere with performance of the PA and CTA tasks in the rat. Furthermore, the data indicate that the dorsolateral periaqueductal gray matter is involved in the performance of the PA task. However, the involvement of the dlPAG in the performance of the CTA appears to be a complex issue. Although effects of muscimol and ibotenic acid on the performance of the tasks point to involvement of the dlPAG, the effects of electrolytic lesions provide also information indicating that other structures in the periaqueductal gray matter can take over for the dlPAG in the performance of the tasks.[New, more effective antidepressants].
